The aim of the research was to quantify the location and movement of the DNA replication factors ORC and MCM in the context of chromatin at the single-molecule level.This was achieved using a combination of optical tweezers and confocal fluorescence microscopy. The position along DNA molecules held in an optical trap of fluorescently labelled H2A, ORC and MCM was tracked over time, and position and motion parameters (e.g. localization and diffusion constants) were studied under different biochemical conditions.
本研究旨在于单分子水平上,量化染色质环境内DNA复制因子ORC与MCM的定位及运动状态。该研究通过光学镊子与共聚焦荧光显微镜的联用技术完成了上述量化目标。研究人员对光阱捕获的DNA分子上经荧光标记的H2A、ORC及MCM的位置进行了实时追踪,并针对不同生化条件下的位置与运动参数(如定位参数与扩散常数)开展了系统性分析。
