Innate lymphoid cells (ILCs) were generated in vitro from CD34+ hematopoietic progenitors derived from umbilical cord blood, and RNA sequencing was performed on the ILC subsets generated to assess global phenotype and compare lineage signatures with each other and to those from ex vivo isolated ILCs. ILCs were generated and cultured in vitro for 30 days, as described in the manuscript, prior to sorting based on surface factors and RNA isolation. CD94+ NK/ILC1 and ILC3 were isolated from the condition of OP9+IL-15, CD94+ NK/ILC1 and CD94- NK/ILC1 were isolated from the condition DL1+IL-15, and CD5- and CD5+ ILC2 were isolated from the condition DL1. A CD161- CD5+ population from the condition DL1 was isolated as an internal, non-ILC, control. From 4 independent donors, 7 populations were sequenced. From 1 independent donor 4 populations were sequenced. Total of 32 samples.
