Sepsis-induced liver injury (SILI) is associated with high mortality and abnormal immune cells activation. Apolipoprotein A5 (APOA5) secreted by liver has an impact on liver immune microenvironment. To explore the function of APOA5 in LPS-induced liver injury and the mechanism of how it affecting liver immune, the mice livers (n=3) were digested with buffer solution including Ca2+ and type I collagenase in vivo. The nonparenchymal cells were acquired through filtration and centrifugation. Finally there are 31852 liver immune cells prepared for scRNA-seq on 10x Genomics. Unsupervised clustering analysis of the data showed ten major clusters based on expressing known markers. It indicated that the depletion of Apoa5 can significantly influence macrophages so that they play a role in protecting the liver from LPS-induced injuries.
