Gas1-CreER+, RT/+ mice were 5X Tmx induced from P26-P30, and obtained at P640 (around 21 months). We enriched the SVZ of 8 aged mice for single-cell RNA sequencing. Aged mice were executed by cervical dislocation, the brain was quickly dissected, washed twice with cold DPBS, and prepared as 1 mm coronal brain lamellae. Carefully and maximally dissect the SVZ tissue under a dissecting microscope (Olympus). Immediately the SVZ tissues were handed over to Novogene (Beijing, China) for tissue digestion and demyelination treatment (Myelin Removal Beads, Miltenyi Biotec, 130-096-433). >10,000 viable cells were used for subsequent library construction and RNA sequencing.
