To investigate the effects of overexpression of amino acid homorepeat (PolyX) fusion proteins on budding yeast cells, we analyzed the transcriptional responses elicited by overexpression of PolyX-fused fluorescent proteins (EGFP-PolyX). Overexpression of multiple PolyX variants induces toxicity in yeast cells, whereas certain PolyX fusion proteins exert beneficial effects. For example, overexpression of a polyisoleucine-fused protein (PolyI-EGFP) causes growth inhibition, indicating high toxicity, whereas overexpression of a polyaspartate-fused protein (PolyD-EGFP) results in growth rate comparable to that of EGFP alone, indicating low toxicity. In contrast, overexpression of a polyglutamate-fused protein (PolyE-EGFP) enhances growth rate relative to EGFP alone, thereby conferring a beneficial effect. This dataset presents RNA-seq results aimed at elucidating the transcriptional responses induced by overexpression of PolyI-, PolyE-, and PolyD-fused fluorescent proteins.
