We previously have found that RIPK3 induces robust anti-viral and inflammatory transcriptional signaling in death-resistant MEFs, however the molecular mechanism that governs this transcription is not well understood. Here we found that RIPK3-depedent transcription is RIPK1 and TRIF dependent in MEFs. Ripk3-2xFv immortalized mouse embryonic fibroblasts (MEFs) lacking MLKL, were treated with siRNA targeting ZBP1, TRIF, RIPK1 and a scramble encoded control. Activatable Ripk3-MEFs were then treated with BB homodimerizer for 4hrs. MEF RNA lysates were then submitted for bulk RNAsequencing.
