The role of Gch1 in the macrophage response to LPS

We used microarrays to detail the global program of gene expression induced in LPS/IFNgamma stimulated macrophages that were deficient in BH4/Gch1 (GCHfl/flTie2cre vs. GCHfl/fl) over a 24 hour timecourse. Bone Marrow-Derived Macrophages (BMDM) were cultured from GCHfl/fl and GCHfl/flTie2cre bone marrow. Differentiated macrophages from 3 animals per genotype were stimulated with LPS in the presence of interferon-gamma and samples were taken for analysis at t=0, 2hrs, 8hr and 24hrs post stimulation.
