Transcriptome analysis of lineage-marked prostate primary tumors and metastases from the NPK mouse model (Nkx3.1CreERT2/+; Ptenflox/flox; KrasLSL-G12D/+; R26R-CAG-LSL-EYFP/+) Male mice of a a predominantly C57BL/6 background were induced to form tumors at 2-3 months of age by administration of tamoxifen using 100 mg/kg (in corn oil) once daily for 4 consecutive days. Following tamoxifen-induction, mice were monitored daily and euthanized when their body condition score was <1.5, or when they experienced body weight loss >20% or signs of distress, such as difficulty breathing or bladder obstruction. Transcriptomic analysis of bulk tissues was done on primary tumors (n=19) and matched macrodissected metastases from lung (n=11), liver (n=5), lymph nodes (n=4), or bone (n=12) from 11 independent mice. RNA was prepared from snap-frozen tissues the using MagMAX-96 total RNA isolation kit and only RNA samples having between >200 ng and 1 ug and with an RNA integrity number (RIN) > 8 were used.
