We performed genome-wide CDK9 ChIP seq experiments in T47D cells only expressing HA Tagged amanitin resistant wild type in comparison to R1810A mutant form of RNAP2. We designed experiment to perform genome wide CDK9 ChIP sequencing in T47D cells expressing HA Tagged amanitin resistant wild type in comparison to R1810A mutant form of RNAP2.We find that PADI2 deiminates R1810 at CTD (C-terminal domian ) of RNAP2 and this is important to regulate the P-TEFb complex association with RNAP2. Therefore in order to investigate precise effect of the RNAP2 R1810A mutantion on CDK9 recruitment, we performed the CDK9 ChIP seq . Objective was to analyse chromatin binding regions of CDK9 in T47D cells expressing wild type (WT) form of RNAP2 was expressed . After 24h of transfections , cells were treated by 6ug/ml amanitin to inhibit endogenous form of RNAP2 .So that we could demonstrate specific CDK9 ChIP seq data after expressing amanitin resistant wild type form of RNAP2 .
