Flow cytometry analysis of beads for <i>Nematostella</i> cell size reference (Fig S6E)

<i>Nematostella</i> cell suspensions were stained with 1μg/ml FxCycle violet DNA dye (Invitrogen) and non-fluorescent polystyrene beads with known diameters between 2.0 um and 15.0 um (F13838, Invitrogen) were added to create a size-reference of FSC-A values. Yellow-green or red fluorescent beads (F8836/F8834, Invitrogen) were used to estimate cell numbers per animal. Flow cytometry was performed a BD LSRFortessa (BD Life Sciences). FSC-A was recorded in particles gated on DNA dye within the expected range of a cell cycle to investigate cell size changes during starvation and the resulting data was analysed using FlowJoV10.8 (BD Life Sciences). .wsp files contain the samples, gating strategy and cell populations used for the analysis in FlowJo.fcs files represent the individual flow cytometry output files that were analysed in the .wsp file<br>
