This project provides a de novo transcriptome dataset of the medicinal plant Schisandra sphenanthera generated to investigate transcriptional responses associated with ecological migration. Plants originating from Luanchuan in Henan Province China were transplanted to Tongdao in Hunan Province China two locations characterized by distinct climatic and soil conditions. Root tissues were collected from plants at the native site and after long term establishment at the migration site. Total RNA was extracted from root samples and sequenced using the Illumina NovaSeq 6000 platform with paired end reads. Clean reads were assembled de novo without a reference genome and unigenes were functionally annotated using public databases. Differential gene expression analysis was conducted to characterize transcriptional reprogramming associated with environmental change with particular focus on secondary metabolism stress response and plant microbe interaction related pathways. This dataset provides a transcriptomic resource for studying environmental adaptation mechanisms in non model medicinal plants and supports integrative analyses of gene expression changes underlying ecological migration in Schisandra sphenanthera.
