Single cell sequencing profiles UGDH mediated TME in liver tumours

Fresh liver tumour samples derived from WT and UgdhcKO mice were dissected and dissociated by minced into 1-2mm3 pieces and digested in collagenase solution for 1 hr. Then the single cell suspension was collected and centrifuged at 500g for 6 mins. Collected single cells were suspended in cold PBS, then counted and controlled for quality. Samples with cell viability more than 95% were processed with a Chromium Controller (10 x Genomics System), captured. Then we executed a library with the Chromium Single Cell 3' Library Kit v2 (10 x Genomics). cDNA libraries were sequenced on a HiSeq 4000 (Illumina) to a minimum depth of 50,000 reads per cell.
