Single cell suspensions of total thymocytes from pooled thymi of 6-week-old BLAB/c mice were enriched with CD138+ cells by MACS. Enriched CD138+ cells and CD138- flow through were FACS sorted into CD19+CD138+ PCs and B220+CD19+ B cells, respectively. PCs and B cells were mixed and single cell sequenced
