The methyltransferase, KMT5A, is suggested as an oncogene in prostate cancer but the mechanisms underlying its oncogenic properties are poorly understood. This study uncovers genes and cellular pathways which are regulated by KMT5A in LNCaP-AI cells, a model of androgen independence. LNCaP-AI cells were grown in steroid depleted media whilst knockdown was performed using 2 independent siKMT5A sequences or a non-silencing sequence over 72 hours. Dihydrotestosterone (DHT) or DMSO vehicle control was then applied for a further 24 hours. RNA was isolated using TRIzol. 4 biological replicates for each sample was carried out. Total of 24 samples.
