Whole-transcriptome of the liver in 15-day-old C3H/HeOuJ and CAST/EiJ mice

Male C3H/HeOuJ and CAST/EiJ mice were treated with diethylnitrosamine to induce liver tumours. We collected liver samples from untreated P15 mice for control experiments. Liver tissue samples were snap frozen in liquid nitrogen and total RNA was extracted using the AllPrep 96 DNA/RNA Kit (Qiagen) according to the manufacturer's instructions. Libraries were prepared using the TruSeq Stranded Total RNA Library Prep Kit with Ribo-Zero Gold (Illumina) and sequenced on an Illumna HiSeq4000 to produce 150bp paired-end reads.本研究使用二乙基亚硝胺（diethylnitrosamine）处理雄性C3H/HeOuJ及CAST/EiJ小鼠，以诱导其形成肝肿瘤。我们收集未经过任何处理的P15小鼠肝脏样本，用于对照实验。将所得肝脏组织样本置于液氮中速冻，并依照制造商的操作指南，使用AllPrep 96 DNA/RNA试剂盒（Qiagen）提取总RNA。随后采用搭载Ribo-Zero Gold的TruSeq链特异性总RNA文库制备试剂盒（Illumina）构建测序文库，并在Illumina HiSeq4000测序平台上完成测序，得到150bp双端读段（paired-end reads）。
