Paramyxoviruses such as human respiratory syncytial virus (HRSV), human metapneumovirus (HMPV) and human parainfluenza virus type 3 (HPIV3) are major worldwide agents of respiratory tract disease, especially in the pediatric population. Basic information on viral replication and host responses is important for understanding and controlling these major pathogens. To this end, we are proposing to use deep-sequencing of cDNA (RNA-seq) to measure the global transcriptional response of human lung epithelial cells to infection by paramyxovirus. Using this technique we will also measure small RNA sequences (< 40 bp) produced during paramyxovirus infection in a human lung epithelial cell line with the hopes of identifying novel small RNAs produced by both the virus and the host cells during the interaction.
