Viral kinetics is the study of viral multiplication in a host (P. operculella larvae) and viral production (amount of granules per harvested larva). A stock solution of PoGV was prepared for each isolate, with a concentration of 109 granules/ml, from which a dilution was made with the factor 4:1 (3 of distilled water: 1 virus) and proceeded to inoculate discs that contained between 800 to 1000 eggs of P. operculella. These filter paper discs were placed in petri dishes, sealing the plates until the larvae hatched. Then, 50 larvae were transferred with the help of a brush to 15 plastic containers of ½ of a liter for each isolation, these containers contained approximately 100g of potato variety peruanita, next they were taken to a growth chamber at 24°C. The evaluations were carried out daily from the sixth day until the 21st day. For this, five larvae of potato tubers were recovered, each larva was placed in a 1.5 ml microtube (eppendorf) and weighed immediately, to avoid weight loss. After, the larvae were crushed in 1 ml of distilled water to count the occlusion bodies. The necessary dilutions were made to perform an optimal count, then 10 μl of the dilution was added to the Neubauer chamber and observed in a phase contrast microscope (Olympus).
