The role of the bone marrow microenvironment for regulating megakaryocyte and platelet function remains incompletely understood. We examined the mechanism by which MSCs can lower platelet activation through communication with megakaryocytes. Overall design: Primary murine wild-type bone marrow CD45-Ter119-CD31-CD51+CD140a+ mscs were cultured together with primary murine wild-type megakaryocytes or as a control, primary murine wild-type megakaryocytes were cultured alone. After 18 hours, megakaryocytes were isolated from cultures, separated from platelets, and analyzed by RNA-seq.
