This dataset provides the underlying source data supporting the findings and figures presented in the manuscript titled "A pathogen-derived small RNA fine-tunes host immunity for chronic parasitism".The dataset is organized into four primary folders corresponding to the figures in the manuscript:Fig.1_data:Numerical Data: Raw values for small RNA length distribution (Fig. 1C) and the TPM expression matrix (Fig.1D).Image Data: Original, uncropped gel image for Fig.1BTransparency: An additional annotation PDF is provided to indicate the specific bands and molecular weight markers.Fig.2_data:Target Prediction (Venn Analysis): Raw gene lists used to generate the Venn diagrams for three miRNAs: OsmiRNA286 (Fig. 2A), OsmiRNA1 (Fig. 2D), and OsmiRNA2 (Fig. 2G). The spreadsheets categorize target genes based on the intersection of three prediction algorithms (miRanda, RNAhybrid, and PITA).Functional Enrichment: Detailed GO (Gene Ontology) and KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment analysis data for the core target genes (intersection of all three software). These files include gene counts, enrichment scores, and p-values corresponding to OsmiRNA286 (Fig. 2B-C), OsmiRNA1 (Fig. 2E-F), and OsmiRNA2 (Fig. 2H-I).Fig.3_data:Luciferase Assay: Raw Firefly and Renilla bioluminescence readings and normalized ratios for Figure 3B.qPCR Data: Raw Ct values and 2−ΔΔCt calculations for Fig. 3C and 3D.Fig.4_data:qPCR Data: Raw numerical data supporting Fig. 4C.Image Data: A sub-folder (Raw_Image_Fig.4D) containing the original, unaltered immunofluorescence (IF) images (DAPI, Cy3, DIC, and Merge channels) at full field-of-view for Fig. 4D.Fig.5_data:Numerical Data: A dedicated folder containing raw qPCR data for Fig. 5B and detailed spore counting spreadsheets for Fig. 5C. The counting files include embedded Excel formulas demonstrating the conversion from raw hemocytometer counts to final concentrations.Image Data: A sub-folder containing original, unaltered microscopy images (captured at 10x magnification) for Fig. 5D.Fig.6_data:Target Prediction: Numerical results of miRanda target site predictions for the attacin 3'UTR across various species (Fig. 6A).Phylogenetic Data: Raw Newick (.nwk) files for the phylogenetic trees constructed using CDS (Conserved) and 3'UTR (Divergent) sequences (Fig. 6B).Nucleotide Diversity: Raw numerical data and calculated Pi (π) values supporting the nucleotide diversity analysis shown in Fig. 6C.Sequence Data: A compiled text file containing the 3'UTR sequences of attacin from multiple species used for sequence alignment and target site conservation analysis (Fig. 6D).Data Standards:Statistical Calculations: All P-values were derived from Student’s t-tests or One-way ANOVA with Tukey’s post-hoc tests as specified in the Excel files.Image Integrity: All provided images are original, high-resolution, and have not undergone any manipulative adjustments.Accession: All analyses are based on raw sequencing data deposited in the Genome Sequence Archive (GSA) under accession number CRA038699.
