Response of unmodified and TGFBR2 knockout tumor infiltrating lymphocytes (TIL) to TGF-B stimulation

TGFBR2 was deleted in ovarian cancer TIL via CRISPR/Cas9 gene editing. Response of unmodified and TGFBR2 knockout TIL to TGF-B stimulation was evaluated via RNA sequencing. Unmodified and TGFBR2 knockout TIL were exposed to 10ng/mL TGF-B1 for 24hrs before collection and RNA-sequencing. TGFBR2 knockout was achieved using two different CRISPR gRNAs: gRNA #3 and gRNA #4. CRISPR gene knockout with each gRNA was performed in triplicate using a single patient TIL sample. Thus, three technical replicates were studied for unmodified TIL (WT), TGFBR2 knockout TIL with gRNA #3, and TGFBR2 knockout TIL with gRNA #4. Each sample was subjected to 24hr exposure to TGF-B1 or vehicle for a total of 18 samples.
