CTCF is recognized for its role in facilitating long-range chromatin interactions and 3D genome organization. In CD8+ T cells responding to chronic infection, CTCF undergoes redistribution to activate the exhausted cytotoxic effector (Tex-eff) program while simultaneously utilizing its insulation function to regulate the fate of precursors to stem-like exhausted T (Tex-prec) cells. hCD2-Cre, Rosa26-lsl-GFP, and CTCF wild-type or floxed P14 CD8+ T cells were adoptively transferred into CD45-disparate recipients followed by infection with LCMV-Clone 13. At 72 hours (day 3) post-infection, GFP+ P14 cells were sorted and purified for pSer5 RNA polymerase II (RNAPII) CUT&RUN analysis.
