AftertheDNAsample(s)was(were)delivered,wedidasamplequality testfirst.Thenweusedthis(those)qualifiedDNA sample(s)toconstructBSlibrary：PurifiedDNAsample,suchasgenomicDNA,BacterialArtificial Chromosomeorlong- lengthPCR productions,is shearedintosmallerfragmentswithadesiredsizebyCovarisS/E210orBioruptorfirstly.Then theoverhangs resultingfromfragmentationareconvertedintobluntendsbyusingT4DNApolymerase,Klenow Fragment andT4PolynucleotideKinase.Afteraddingan‘A’ basetothe3'endofthebluntphosphorylatedDNAfragments,adapters areligatedtotheendsoftheDNAfragments.Thedesiredfragments canbepurifiedthoughgel-electrophoresis,then selectivelyenrichedandamplifiedbyPCR.Theindex tagcouldbeintroducedintotheadapteratthePCR stageas appropriateandwedidalibraryquality test.Atlast,thequalifiedBSlibrarywouldbeusedforsequencing.
