Development of a novel methodology to identify single-strand break sites ('nicks') in DNA. Nick detection is based on consecutive nick translations with dPTP and dKTP, synthetic degenerate nucleotides, and then with biotin-dUTP which provide mutational and enrichment signals in the final sequence data. DENT-seq (formerly NickSeq) is utilized to identify off-target nicking by Cas9 nickase in human genomic DNA.
