LC–MS metabolite profiling data for methionyl-tRNA synthetase knockdown in Mycobacterium abscessus

This dataset contains LC–MS-based metabolite profiling data generated from Mycobacterium abscessus MetRS_KD and PLJR962 control samples, with four biological replicates per group. The deposited files include vendor raw LC–MS data, sample and assay metadata, a detailed method description, and processed metabolite abundance tables. Metabolites were separated using a SeQuant ZIC-HILIC column (100 mm × 2.1 mm, 3.5 µm) on a UPLC system coupled to a Q Exactive Plus mass spectrometer. Water containing 10 mM ammonium formate and 90% acetonitrile containing 10 mM ammonium formate were used as mobile phases. Data were acquired in both positive- and negative-ion modes using data-dependent acquisition, consisting of one full MS scan followed by five data-dependent MS/MS scans. Raw LC–MS data were processed using TraceFinder against a predefined mycobacterial metabolite list containing 224 metabolites of interest. Metabolite annotation and quantification were based on available LC–MS characteristics, including accurate mass, retention time, and MS/MS information. Authentic reference standards and/or previously established analytical parameters were available for a subset of metabolites. These data are associated with the study “Methionyl-tRNA synthetase inhibition exposes redox–bioenergetic and collateral drug vulnerabilities in mycobacteria.
