Ruminants have a special digestive system. The complex forestomach includes the rumen, reticulum and omasum. The rumen is the most important digestive system and provides 70% of the animal's daily energy needs by decomposing fiber in the feed. Organoids are an efficient and reliable research model. In this study, we established rumen epithelial organoids and explored the transcriptional dynamics of organoids during development. We found that the Hippo signaling pathway plays a key role in the development of rumen epithelial organoids. We used Verteporfin and GA-017 to inhibit and activate YAP respectively and analyzed the effects of inhibition/activation of YAP on the transcriptional profile of rumen epithelial organoids.
