Total cellular DNA contains only 1 - 10% of chloroplast DNA. The transfer of plastid DNA fragment to nuclear and mitochondrial genome are frequency found in plants. In rice, such transfers occurs more abundant compared with other higher plants and those sequence affect downstream analysis of Next-generation sequencing. Therefore, the isolation of high purity chloroplast DNA can provide next-generation sequencing of rice chloroplast genome with high resolution, high accuracy and cost-efficiency.
