The 2’-O-methylation (2’-O-Me) of ribosomal RNA (rRNA) shows plasticity potentially associated with specific cell phenotypes. We used RiboMeth-seq profiling to reveal senescence- and proliferation-specific 2’-O-Me patterns in stress induced premature senescent (SIPS) human dermal fibroblasts. Primary human dermal fibroblasts (HDF) from three healthy donors were kept sub-confluent (= proliferating, P), confluent (=quiescent, Q) or senescence was induced by hydrogen peroxide (=stress-induced premature senescence, SIPS). Duplicates of each cell state from each donor were analyzed by RiboMeth-seq.
