Diarrhea occurs in children and toddlers in the world, especially those aged five years and under, and is the second leading cause of death with a total of 370,000 deaths per year based on WHO data in 2019. Diarrhea cases in children still account for the largest proportion of causes of death in Indonesia. Based on data obtained from the Directorate General of Public Health, Ministry of Health of the Republic of Indonesia in 2020, around 314 cases of diarrhea were recorded or 10.7% contributed to deaths out of a total of 2,927 deaths among children under five in Indonesia. Bacterial, viral and fungal pathogenic agents can cause diarrhea. Enteric bacterial diarrheal disease caused by botulism, Campylobacter gastroenteritis, cholera, Escherichia coli gastroenteritis, Salmonellosis, Klebsiella oxytoca, Shigellosis (Shigella spp.), and food poisoning Staphylococcal is the most common type of bacteria found to cause moderate to severe diarrhea. Gold standard pathogen microbiome detection methods include PCR and culture. However, the disadvantage is that the time required for culture is longer, with a minimal number of pathogenic agents. In this research, we will map the diversity of pathogenic agents (bacteria) that cause diarrhea in children aged 2-5 years using the 16s rRNA metagenomic sequencing method.
