A map of the rubisco biochemical landscape

Deep mutational scanning study on Form II rubisco from Rhodospirillum rubrum. A library of most single mutants of rubisco were assayed in a modified E. coli strain with growth coupled to the carboxylation activity of rubisco. Challenging the library with different concentrations of CO2, we computed the Michaelis constants many mutants including several with improved CO2 affinities, which were then confirmed biochemically.
