Double mutants of the genes encoding chromomethylases in maize (zmet2 and zmet5) are nonviable, as are double mutants of the genes encoding DDM1-type nucleosome remodelers (chr101 and chr106). Despite being nonviable, both double mutants produce embryos and endosperm. We prepared tissue from developing embryos, developing endosperm, and mature endosperm from mutants to investigate their effects on DNA methylation and small RNAs.
