This project includes raw sequencing data from genome-wide transposon sequencing (Tn-seq) experiments performed in Aggregatibacter actinomycetemcomitans (Aa), a major periodontal pathogen. The goal of the study was to identify genetic determinants that support Aa fitness under anoxic conditions using trimethylamine N-oxide (TMAO), its preferred anaerobic electron acceptor and a common dietary metabolite. Genome-wide transposon mutant libraries were cultivated under the following conditions: (1) Input: Frozen transposon mutant libraries revived in nutrient-rich media, representing baseline mutant abundance; (2) Control: TSBYEP (tryptic soy broth supplemented with yeast extract and peptone) incubated under anoxic conditions without TMAO; (3) TMAO: TSBYEP supplemented with 50 mM TMAO and incubated under anoxic conditions. Sequencing of the libraries was performed to quantify mutant abundance and assess gene fitness across conditions. These data provide insights into the genetic requirements for anaerobic respiration and survival of A. actinomycetemcomitans in the presence of TMAO.
