Low Dose Pro-resolving Mediators Reset the Resolution Response to Microbial Inflammation

With murine zymosan peritonitis, repetitive dosing of a SPM panel (RvD1, RvD2, RvD5, MaR1 and RvE2 at 1 ng/mouse, i.p.) was given to mice, followed by zymosan challenge to initiate peritonitis. Leukocyte composition and resolution indices were calculated. RNA-sequencing was carried out Overall design: Mice were given "treatment" i.e., a SPM panel (RvD1, RvD2, RvD5, MaR1 and RvE2, 0.1 ng each/mouse, i.p.) or "vehicle" controls on days 0, 2, 5, 7 and 9. On day 12, zymosan A (1 mg/mouse, i.p.) was given together with either the SPM panel or vehicle. Peritoneal exudates were collected at 12, 24 and 48h, and leukocyte subtypes were assessed using flow cytometry. Aliquots of exudate leukocytes collected from 12h time point were used for RNA-seq.
