Chemokine Ligand 2 Expressing Fibroblast Recruitment of Macrophages Enhanced in Loeys-Dietz Syndrome Aortic Root Aneurysm Formation [human]

Loeys-Dietz Syndrome (LDS), caused by a spectrum of mutations in the TGF-ß signaling cascade, leads to aggressive thoracic aneurysms. Given the ubiquitous nature of TGF-ß signaling machinery, the exact cell type(s) responsible for initiation of LDS pathogenesis is poorly understood. Overall design: Aortic root and ascending aortic tissue from 5 LDS (Loeys-Dietz Syndrome) and 2 Donor control patients was subjected to tissue digestion and single-cell RNA sequencing. Aortic root and ascending aorta samples were processed as individual libraries from each patient, yielding 12 samples total.
