The goal of this experiment was to investigate the effects of Arabidopsis MYB transcription factor (TF) AtMYB12 on flavonoid biosynthesis in Glycyrrhiza inflata hairy roots. Eight-day-old G. inflata seedlings were used for hairy root generation. Through overexpression of AtMYB12 in G. inflata hairy roots, in comparison with the empty vector (EV) control lines, the flavonoids accumulation and expression of flavonoid biosynthetic genes were studied. Three AtMYB12 overexpression hairy root lines (AtMYB12-OX) and three EV control lines were used. These hairy root lines were cultured in 125 ml flasks each containing 10 ml MS liquid medium on an orbital shaker at 100 rpm at 25 C. The hairy roots clones were routinely sub-cultured every two weeks and harvested after 2 months for RNA-sequencing. This yielded a total of 6 samples - three EV control lines (EV-1, EV-2 and EV-3) and three AtMYB12 overexpression lines (AtMYB12-OX-1, AtMYB12-OX-2 and AtMYB12-OX-3).
