In this study, we established 2 EGFR-mutant SCLC cell lines from lung adenocarcinoma patients after failed EGFR-TKI treatment. These 2 EGFR-mutant SCLC cell lines displayed 2 different phenotypes: suspensive and adherent. We used microarrays to identify the global gene alterations and molecular basis of EGFR-mutant SCLC cell lines. The patient-derived EGFR-mutant SCLC cell lines CLH and CLH5 were established in F medium, and after several passages were adapted to RPMI medium. Both CLH and CLH5 cell lines different phenotypes: suspensive cells (CLH-S and CLH5-S) and adherent cells (CLH-A and CLH5-A). The gene expression profiling of these 4 cell lines were performed by microarray analysis. We tried to compare the gene expression profiles of adherent cells with suspensive cells to identify the specific pathways include tumor growth, metastasis and drug resistance.
