资源简介:

Dataset description This dataset collection accompanies the publication “Hyper Lightsheet for live three-dimensional fluorescence imaging under hypergravity” by Pampaloni et al. The collection contains the underlying data used for the plastid-tracking analysis in Arabidopsis thaliana and for the analysis of the acceleration-induced specimen displacement during hypergravity imaging. The plastid-tracking dataset contains time-lapse fluorescence microscopy data of Arabidopsis thaliana roots expressing the pEGAD-X plastid marker, together with the data used to quantify plastid trajectories under hypergravity. The acceleration-induced displacement dataset (file Data Root migration 8 bit.tiff) contains a time series of an Arabidopsis thaliana pEGAD-X root tip acquired while the centrifuge acceleration was progressively increased. Increasing acceleration resulted in a systematic displacement of the specimen in the x, y, and z directions. The measured displacement was used to quantify acceleration-dependent drift and to align a representative optical plane through the root, thereby illustrating how specimen displacement can be corrected during hypergravity imaging. Imaging was conducted using a Carl Zeiss W N-Achroplan 20x/0.5 water-dipping objective lens. Excitation was provided by a 488 nm laser line/diode at maximum power. The fluorescence signal emitted by the marker was collected using a 440/521/607/700 HC Quadband emission filter (AHF Analysentechnik, Germany, F72-888). These datasets provide the underlying experimental data for the corresponding quantitative analyses and figures presented in the publication.
