We used an adenine base editor to target the translation start site and mRNA splicing site of Camk2d in order to knock out CaMKIId. We found that editing the 5' splice site of intron 7 can lead to premature translation termination, effectively knocking out CaMKIId. Overall design: In order to investigate the impact of editing the 5' splice site of Camk2d intron 7 using an adenine base editor on mRNA expression, we designated the edited mice as the experimental group and the unedited mice as the control group. We compared the changes in RNA expression in the hearts of both groups using RNA-seq.
