The purpose of this experiment was to evaluate the human host cellular response to wild-type Human coronavirus strain 229E (HCoV-229E) infection in the presence and absence of macrophages. Sample data was obtained for mock and infected (MOI 3) primary human airway epithelial cells with and without macrophages and grown in air-liquid interface conditions, processed for RNA sequencing (RNA-Seq) expression analysis. Overall design: Mock infected and human coronavirus 229E infected samples in the presence or absence of macrophages were collected in primary human airway epithelial cells at 24 hours post infection. Cultures were first washed, inoculated with human coronavirus 229E for 2 hours and the inoculum removed and the cultures washed twice. Freshly harvested primary human macrophages (isolated from PBMC) were then incubated on the apical surface for 4 hours prior to removal and two additional washes of the cultures.
