Normalized cDNA library was constructed from mRNA that was extracted from various castes/developmental stages of Hodotermopsis sjoestedti collected from Yaku-shima Island, Kagoshima, Japan. We employed 454 FLX Titanium system for sequencing the normalized cDNA library, assembled 454 reads with Newbler v2.5.3, and created a EST contig set. To identify genes that respond to a juvenile hormone analog (JHA) application in pseudergates of H. sjoestedti, we carried out JHA treatments on pseudoergates and Suppressive Subtractive Hybridization (SSH). We determined nucleic acid sequences of gene fragments generated from the SSH, and performed similarity searches for those fragments in the 454 EST contig set, using the blastx and tblastx algorithms.
