Studies have demonstrated that mild hypothermia therapy exerts a positive protective effect on the blood-brain barrier (BBB) following ischemic stroke; however, there remains a paucity of research elucidating its underlying molecular protective mechanisms. To further investigate the molecular pathways through which mild hypothermia protects the BBB in ischemic stroke, as well as its reprogramming effect on BBB lipid metabolism, we established an in vitro oxygen-glucose deprivation/reperfusion (OGD/R) model using the mouse brain microvascular endothelial cell line bEnd.3. The experimental design included a control group, a model group and a mild hypothermia intervention group. Subsequently, transcriptomic and lipidomic analyses were performed on the harvested cells from each group.
