This whole genome sequencing project validates the site-specific integration and in-frame fusion of mNeonGreen::T2A (mNG::T2A) tagging cassettes in Trypanosoma brucei. The study includes five engineered cell lines where different genes (ESAG3, ESAG7, and Hsp70) were tagged with either mNG::T2A or mNG::6xTy cassettes, along with an unmodified wild-type control. Sequencing was performed using paired-end sequencing at 30X genome coverage to confirm correct integration, proper reading frame maintenance, and absence of mutations at the target loci. This dataset supports the development of an efficient endogenous protein tagging system in T. brucei for both secretory and cytosolic proteins.
