We are investigating the functional role of a gene that encodes a cell surface receptor belonging to the class of adhesion G-protein coupled receptors. The Sanger Institute mouse genetics programme showed that female (but not male) homozygous mutant mice were completely infertile. Our subsequent research has shown that the fertility phenotype does not appear to be due to failures in either gamete development, recognition or embryo implantation, but rather in the transport of ovulated eggs through the oviduct. We have shown that the protein product is located on the epithelial surface of the oviduct which therefore suggests active signalling through this receptor creates a permissive environment for egg transport, although the mechanism by which this might occur is a mystery. To gain insights into the mechanistic basis of this effect we will perform RNAseq analysis in the oviductal epithelium from knock out and control wild-type mice following ovulation.
