G-quadruplexes (G4s) are non-helical secondary structures that can fold in vivo in RNA from stacked quartet of guanine bases. In human cells, they can affect translation, transcript processing and stability of RNA.Putative G4 sequences are predicted in the transcriptome of the malaria parasite Plasmodium falciparum, despite a very highly A/T-biased genome with unusually few guanine-rich sequences.Here, we applied the rG4-seq (RNA G-quadruplex sequencing) technique on parasite samples for transcriptome-wide probing of RNA G4 structures. A comprehensive landscape of RNA G4s enables exploration of connections between the RNA structure and parasite biology.
