The ISPpu10 transposon, a member of the IS110 family, utilizes a bridge RNA (bRNA) to mediate programmable DNA recombination. In this project, we repurposed the ISPpu10 system to develop a novel, highly specific tool for large-scale genome editing. This BioProject contains third-generation long-read sequencing data generated to validate the successful, targeted integration of a donor plasmid into the bacterial genome (e.g., Pseudomonas putida KT2440). Long-read sequencing was performed to verify the precise insertion site and assess the structural integrity of the integrated payload. These data correspond to Figure 5 of our manuscript and demonstrate the accuracy and feasibility of the ISPpu10-based system for programmable genomic insertions.
