The heterogeneity of antigen-specific naïve CD8 T cells has not yet been fully elucidated. We found that long-lived naïve CD8 T cells, incubated in naïve B6 mice for more than 30 days, generate significantly higher numbers of effector and memory CD8 T cells after lymphocytic choriomeningitis virus (LCMV)-Armstrong infection compared to freshly isolated naive CD8 T cells. To examine the mechanism of why long-lived naive CD8 T cells exhibit superior responses to infection, we compared transcriptional profiles between effector CD8 T cells derived from long-lived and freshly isolated naive CD8 T cells. Overall design: RNA-seq profiles of effector P14 T cell receptor transgenic CD8 T cells derived from freshly isolated and long-lived P14 CD8 T cells on Day 5 post-LCMV-Armstrong infection.
