FRAP analysis of highly mobile intracellular structures, such as IAV inclusions, poses significant challenges due to their substantial "bulk-drift". Tracking them with traditional FRAP becomes nearly impossible. Nevertheless, the fluorescence loss after photoactivation (FRAPh) technique offers a solution. It involves pulse photoactivation of individual or pools of viral inclusions at specific sites, allowing tracking over time at photoactivated and non-photoactivated regions regardless of "bulk-drift". This enables the quantification of interaction dynamics of intracellular structures even in the presence of bulk drift, providing valuable insights into their behaviour.
