Streptomyces species are characterized as gram-positive bacteria with GC-rich linear genomes and serve as a dominant reservoir for clinically and industrially important secondary metabolites with antibiotic, anthelmintic, antifungal and even immunosuppressive activities. Genome mining of Streptomyces revealed that each Streptomyces species typically encodes 20 – 50 secondary metabolite biosynthetic gene clusters (smBGCs), further emphasizing their importance. Despite its enormous genomic potential, most of the smBGCs are uncharacterized in terms of their products and regulations due to the silent nature of the smBGCs in laboratory culture condition. To link the Streptomyces' genomic potential to practical applications, understanding the complex regulation and identifying the underlying regulatory elements responsible for secondary metabolism are essential. Here we describe Next-Generation Sequencing techniques applied to two Streptomyces species, including Streptomyces coelicolor and Streptomyces griseus, to reveal the boundaries of transcripts in genome scale, providing fundamental resources to broaden our understanding on Streptomyces' complex genetic regulations and elevating their potential.
