pyrosequences of microbiota in diabetic mice and human

To analyze responsible alterations in intestinal microbiota for protective ability of infection with an intestinal nematode against drug-induced diabetes in mice, fecal DNA was collected from streptozotocin-treated mice with or without infection with Heligmosomoides polygyrus. In human, to compare mirobiota between patients with type 1 diabetes and healthy volunteers, fecal samples were also collected from 15 patients and 16 volunteers, and DNA was extracted. The 16S rRNA DNA was amplified using the universal primers and pyrosequenced by the next generation sequencers. The amplicon reads were processed using QIIME and Mothur. Operational Taxonomic Units (OTUs) and assigned a taxonomy using the Ribosomal Database Project database.
