ChIP-Seq was carried out to map the genome-wide chromatin occupancy of ZNF148 in a basal subtype triple negative breast cancer cell-line, MDA-MB-231. Overall design: MDA-MB-231 cells stably expressing the bacterial biotin ligase Bir A were stably transfected with recombinant forms of ZNF148 (Bio-ZNF148), containing an amino terminal BirA recognition/biotin acceptor motif. Clones that expressed the recombinant proteins at close to the same levels as the endogenous proteins were selected. The parental MDA-MB-231 cells expressing BirA alone were included as controls. The BirA and Bio-ZNF148 expressing MDA-MB-231 cells were then subjected to streptavidin-based ChIP-sequencing.
