To screen the potential targets of Salmonella 3’ UTR derived sRNA FadZ, we compared global mRNA profiles in FadZ-deficient and -pulse expressed strains. The 40-nt FadZ sequences was cloned onto a plasmid downstream of the pBAD promoter and expression was induced by L-arabinose for 10-min in LB broth. Transcriptomic profiles were compared with strain carrying the empty control plasmid. Salmonella FadZ-deficient strain (ΔfadBA- ΔFadZ) carrying either empty plasmid pBAD or FadZ-overexpressed plasmid pBAD-FAdZ, cultivated in LB broth to OD600~1.0 and then induced by 0.2% induced by L-arabinose for 10-min; RNA were collected by hot-phenol method and expression difference were compared between strain with empty and FadZ-expressed plasmids.
